NAD⁺ is a coenzyme involved in hundreds of processes in the body, including:

  • DNA repair – repairing damage to genetic material
  • Cell communication – transmitting signals that determine how cells behave
  • Defense against oxidative stress – protecting cells from free radicals

How nicotinamide riboside works: how exactly does this supplement work?

When you take an NR supplement, it enters your bloodstream and is converted into NAD⁺ through a number of biochemical steps. This conversion takes place via the so-called salvage pathway, a kind of recycling route in your cells that ensures NAD⁺ stores are replenished.

Once formed, NAD⁺ is used in the mitochondria—the energy factories in your cells. There, it helps produce ATP, the universal energy unit needed for almost everything your body does, from muscle contraction to brain activity⁶.

In addition, NAD⁺ plays a role in activating sirtuins, enzymes involved in DNA repair, reducing inflammation, and cell renewal. These sirtuins are often mentioned in research in connection with healthy aging⁵.

How nicotinamide riboside works in supplements

In supplement form, NR is often available in 150 mg capsules. The usual dosage is between 150 and 300 mg per day, with many people choosing 2 capsules spread throughout the day for a steady NAD⁺ replenishment.

Why not take NAD⁺ directly?

NAD⁺ supplements do exist as well, but this substance is not absorbed well in the intestines. NR has a smaller structure and can therefore pass through cell membranes more easily, after which it is converted into NAD⁺ inside the cell itself.

Combinations with other substances

Some users combine NR with resveratrol and TMG.

  • Resveratrol is associated in research with activation of sirtuins.
  • TMG (trimethylglycine) supports methylation processes, which are important for converting NR into NAD⁺

How quickly do you notice anything from NR?

This varies per person. In studies, higher NAD⁺ levels are often measured within 2 to 4 weeks. Whether you also feel subjectively more energy depends on factors such as your lifestyle, age, diet, and overall health.
